Cassette mutagenesis is a type of site-directed mutagenesis that replaces a fragment of target DNA with a short, double-stranded oligonucleotide sequence called a gene cassette. Specificity is achieved through complementary restriction enzyme digest ends on the target DNA and cassette. A single cassette can contain multiple mutations, and the method does not involve primer extension by DNA polymerase.
